AI-Enhanced Single-Particle Tracking – SptPALM
Mapping Single-Molecule Dynamics
Single-Particle Tracking Photoactivated Localization Microscopy (SptPALM) is designed to map single-molecule dynamics.
It uses a photo-activable protein called mEOS (from coral) to track proteins of interest.
This technique has brought unprecedented insights into molecule dynamics, from neurons to plant cells.
SptPALM inventors were awarded the Nobel Prize in 2014.
The Challenge: mEOS Blinking
One difficulty with mEOS is that it blinks, and this blinking is relative to laser power used for activation.
Biologists face a tradeoff between:
Signal-to-noise ratio (improves with higher laser power)
Tracking time of the protein-mEOS fusion (longer at lower laser power)
AI-Powered Solution by BionomeeX
BionomeeX, in collaboration with CNRS researchers, developed an AI-based algorithm to enhance SptPALM imaging under very low laser conditions.
The algorithm processes videos taken at low laser power
Achieves particle track recovery 3× longer than traditional high-laser-power imaging
Maintains high accuracy while reducing phototoxicity and photobleaching
